Woman: When our parents were away I was expected to look after the affairs.
Her friend: That's because you were the oldest?
Her friend: That's because you were the eldest?

It's more natural to say 'I was expected to look after the house and my sisters'. Either 'eldest' or 'oldest' can be used in this context.
