I read #1 in a book on English preposition.
so I am curious whether both #1 and #2 are grammatical or not.
or wheather #1 and #2 have same meaning or not.

#2- When does the next bus for Oxford come? When does the bus that will go to Oxford arrive in London? eg maybe the bus started its journey in Manchester.

I think #1 means "A man is waiting for that the bus for Oxford will leave.from the bus stop. and asking someone
"When does the next bus for Oxford leave for Oxford?".but we don't know whether he is on the bus or not.
If he is on the bus

There is also the possibility that the person is waiting for the arrival of the bus (from, for example Manchester) that will then head off to Oxford because he is meeting a friend arriving on that bus. Clearly he is interestested only in when the bus arrives/comes, especially if the bus has a ten-minute toilet break at this bus stop.
