Today I played the best way I have ever played 'in' my life.

Is of possible instead of in?

Yes, as a statement of life experience. I see nothing wrong with 'in my life', but as Clive says, the idiom uses 'of'.

Does the word 'of' work? Does not sound good in that sentence. But in the one that Clive provided sounds like both are possible but of course the idiom is Of.
