Jerusalem had been totally destroyed six centuries earlier by Nebuchadnezzar, King of Babylon. Within fifty years of that first destruction, the Temple was rebuilt and the Jews returned. (Jerusalem : the biography)

Hi. Does the underlined part mean "Suppose the first destruction is in year 1, both the time of the beginning of the rebuilding and time of the completing of the rebuilding can be any time between year 1 and year 50, but not after year 50", right?

More or less, yes. Strictly speaking, it could be completed in year 51; e.g. if destroyed in December of year 1, then March of year 51 is still within 50 years.
