'what the matter is' is the object of the verb 'know'. In a noun clause beginning with a question word, the subject precedes the verb. Do not use question word order in a noun clause.

The question is not clear.
If you meant "right" from a grammatical point of view, then no, it isn't correct. The word "people" does not belong in that sentence. And, if "most" girls are prostitutes, then it does not make any sense to say that "a lot" are not. After you've mentioned "most", one would supposed that not "a lot" are left.

Now, I don't agree with Jim that your sentence sounds offensive... simply because, most probably, no one in their senses would take a sentence of this type, posted out of the blue by an anonymous poster, seriously.

In any case, next time you wish to say soemthing like that about women, you can try:
"Most girls are prostitutes; few are not."

Be careful, however, to use it in the appropriate context, because we don't have an emergency room in the forums! ~chuckles~
