Neither are exactly correct. They are both very close but you would need to say 'states' and 'the USA'. So to make the sentences correct you would say "How many states are there in the USA" or "How many states is there in the USA"

You could also simply say - "how many states in the USA?"

I competely missed that the word "state" was singular in the orignal sentence. Sorry about that.
