Which sentence is correct ?

Not in queue or Not on queue
eg : Are you on/in the queue ?

It's highly regional, at least in the US (if you replace "queue" with "line" since we don't really use "queue"). Either is acceptable. " but I'm aware that such an expression comes to some of us as naturally as breathing.

It's highly regional, at least in the US (if you replace "queue" with "line" since we don't really use "queue"). Either is acceptable. I personally would never say "Are you on line?" but I'm aware that such an expression comes to some of us as naturally as breathing.

As an object of a preposition "...in the queue" is correct, for example: We had to wait in a queue to see the movie.
Can also be used alone, as in: The queue at the bank was too long, so I decided not to wait.
The word cue is used with on. For example: The actors were right on cue with their lines.
