Which is the correct sentence?

They practise aikido in a 170-tatami mat hall.

They practise aikido in a 170 tatami mat-hall.

They practise aikido in a 170-tatami-mat hall.

Which is the correct sentence? If none of them is correct, how do I punctuate the sentence?

Since a tatami is a mat it is redundant to say tatami mat . If you omit the mat , you would have a 170-tatami hall , which would make clear that there are 170 tatamis. If you wish to leave the ma t in there I would recommend your third sentence.
