I lean a bit toward "in."
The meanings seem different to me.

Recalling the famous bridge, "Let us be happy in our work."

I'm thinking of the sense in which one is happily situated in his profession. (engrossed in his work/profession)

Normally, with would be used, as in content with one's profession. I took that to be the aim of the OP.
