What's up

Hello everyone. I have a question.

Regarding the following dialogue:

John: I don't know why, but I've been feeling very tired recently.

Alice: What's wrong? Are you getting enough sleep?

I know that Alice can say both "What's wrong (with you)?" and "What's the matter (with you)?" here, but I'm not sure if she can say "Whats' up?" in this situation. It sounds strange to me, but I don't have enough confidence.
