What is the meaning of are/is/am not in interrogative form?

Do we say aren't when we are sure

Or when we are asking for something(unsure)

For example I saw a photo of my friend

And I said to myself isn't it Jacob?

(I am sure it is he)

Or My friend brought me mushrooms

I asked him Aren't these poisonous?

(I am asking my friend about it Neither i know that they are poisonous Nor they look poisonous so i am not sure)
