I don't know the context of that answer, Jr Danny, but I find it very strange to say "YES, I do NOT have something"...

1. Person 1: " I think you don't have brains"

Person 2: " Yes, I don't have brains."

2. Person 1: " I think you have brains" or "Do you have brains?"

Person 2: No, I don't have brains"

Yes and No are used to show agreement/disagreement with

I still find the first dialogue a bit strange, but that may be personal.
