Using "S" at the end of word with does

"The man looks like he's tired"

vs

"The man does look like he's tired"

I'm asking if when we use "does" in the sentence, we don't use plural anymore?

There is no plural in your first sentence.
The "third-person singular" (he, she, it, the man, an elephant, etc.) takes an S ending in regular verbs. It is the singular form, NOT the plural.

The second sentence was correct as written.

When you have a form of "to do" (does he, did he, he does, he did, doesn't he, didn't he), the main verb stays in its bare form ("look").

Right. Except on a verb, the "s" doesn't mean it's plural, so you should state it like this.

When we use "does" in a verb phrase, we don't use "s" on the other verbs in the verb phrase.
