You have to put the correct stress on the appropriate words. When said, it has to sound like a question. That's the beauty of phone converstions, the fact that you can not see facial expressions, more effort has to be made with the voice to convey the right message.

I thought you meant that 'is someone available' may have a different meaning if said with improper intonation.
