Usage of I and me

Which one of these sentences is correct?

There are people plotting against Joe and I.

or

There are people plotting against Joe and me.

In singular form you would say, "There are people plotting against me." I'm under the impression that Joe and me is the correct usage.

Your impression is correct: You would say "against me" so the correct form is "Joe and me."

Too many people were frightened away from using "so-and-so and me" because they used it as the subject when they were children and were told over and over "It's 'Joe and I'!" that
