Up and over

In a movie, I heard a person give directions to a place as follows :
"Go 6 blocks up and 3 over." What does this mean ?

Presumably it means six blocks in one direction, then three blocks in a direction at right angles (or roughly at right angles) to the first direction. The initial direction, "up", and the direction of "over" (whether left or right) are context-dependent and cannot be determined just from this quote.
