Two "maybe" questions

1. How do we know when to use "would" and "will" in sentences? I am asking that because sometimes I think they could be used interchangeably without changing the overall meaning.

Would you do this?

Will you do this?

2. How come in these sentences, one could use a comma and one apparently couldn't?

The burglar got into Jane's flat using builders' ladders.

We made the cake from scratch, using no prepared ingredients.
