Dear

"I couldn't see Mr. Sam, who was too/so busy today."

Is it also right if I use "so" instead of "too"?

Only in informal English. Formally, 'so' requires a subsequent 'that', hence a different sentence: Sam was so busy today that I couldn't see him.
