Too

Do we only say 'too' as a reply back to what someone has said to us?

Example:

Elena: Thank you for protecting me.
Edward: Thank you, too, for taking care of me.

Or can we also say it as something in addition to what we've said earlier?

Example:
Elena talking to Edward and Jacob.

Elena: Thank you, Edward.
Elena: Thank you, too, Jacob.

Is the comma (,) required before the word 'too' like 'I love you, too'?
