The word 'Besides'

You there !

Please tell me, should I use or not a comma after the word "besides" ?

For example: They are going to spend the vacation there. Besides (or Besides , ?) they will improve their knowledges (is it correct, their knowledges ?) in that language.

By the way do we say: They have lots of knowledge (or) they have knowledges?
