The use of 'a' and 'an'

Is it grammatically correct to use 'a' before a word starting with a vowel when it doesn't sound correct to use 'an'?

E.g. Can I say 'a uniquely' rather than 'an uniquely'

Yes, as uniquely starts by the sound /ju/
