What would be the answer after nice to meet you?
Would it be you too or me too?

It's not a question, so it doesn't need an answer. Just say the same thing.

When you meet someone say Nice to meet you.
You can add "too" if you aren't the first to say it, or if you are the first to say it, the other person may add "too".

'How do you do?' sounds rather formal/old-fashioned to me these days.
