Can I us the Thank and best regards, (with comma) as my ending? or Thank you and best regards. (period) followed by Sincerely yours, ?

A comma, nt a period . Best regards is an ending for informal letters. eg Thank you and best regards, Sincerely yours or Yours sincerely is an ending for more formal letters.
