SUPERLATIVE "OF" + "IN"

Hello everyone,

I have a question regarding "of" + "in" with superlative. I know that "in" appeals to singular nouns, while "of" to plural ones. For example:

He is the best in town.
He is the best of all.

However, are there any exceptions ? What would be better to say ?

"That's the best part of the job."
or
"That's the best part in the job."
or
"That's the best part with the job."

Grateful to all of you.

Simply other situations:
Now we come to the highlight of our day.
It is one of the perqs of this jo
