Subject agreement

What is the rule on which I can choose the right verb? I find difficult to determine whether the verb refers back to a singular or plural subject. Here some question to clearify it much more:

One of the people who (know/knows) a lot .

It's you who (is /are) reponsible for students.

It (is /are) you the director.

Ungrammatical no matter which choice you make.
