One of the forum gentlemans answered my question like above.

I don't know why 'with' 3 is necessary, and why 2 without 'with' is weird.

And please tell me if there are similar cases like a preposition supplement in 3.

'With' or a some other way of complementing is necessary with the verb 'help' and 'something' as the noun object, that is all. That is the nature of the verb. Try them without 'there is':

3: Can I help you with something?

2: (X) Can I help you something?

Other verbs take other prepositions, or have optional prepositions, or no prepositions, and
