Is the sentence correct? Can I use 'show up' this way?

John didn't show up for work last week because he was on leave.

Well... I'm not sure I agree with this usage. If he was on leave, he was not supposed to be at work. You don't expect him to be there.

Saying "He didn't show up" sounds like he was supposed to be there and was not.

A small further comment about 'on leave'.

I don't hear this expression a lot in N. america.
What I hear is eg
He's on vacation.
