Please tell me if on is necessary here.

We are planning to have English classes in the afternoon between 1.50 and 2.50 pm on two days of the week.

The way it is written it does need the 'on'. 50 pm, two days of the week. but using 'on' is better:)

The way it is written it does need the 'on'.
You can also try using punctuation:
We are planning to have English classes in the afternoon between 1.50 and 2.50 pm, two days of the week.

but using 'on' is better:)
