I know you did not come to the office last week, but you said earlier you must come to the office.

That is grammatical. You might want to replace "but" with "although."

I would also say "had to" instead of "must."

Is your second part, about the vacation, linked to the first? What is the person became seriously ill or suffered a family emergency. You don't want to imply that the person was on vacation if you don't know for sure that it's true.
