Which is the most natural or correct?

- My room is the second to/from/before last down the hall.

I wouldn't use any of them. They are at least not necessary and some are seriously problematic.

'second from last' and 'second before last'

That may be so in Canadian or U.S. English. But here in old blighty we normally say:
Last
Last but one OR penultimate
Second before last

There is a longer world for second before last but I forget it off hand.
