1. Do I say 'Sally asked me to complete 1 exercise of Essentials of Writing' or 'Sally asked me to complete 1 exercise in Essentials of Writing' ?
2. Should I use the word 'one' instead of the Arabic numeral?
3. Should I put inverted commas around Essentials of Writing? What about italicising the words?

What do you mean precisely by an 'inverted comma'? If you mean the marks that I have placed around 'inverted comma', then those serve as informal quotation marks, as quotation marks within quotation marks, and as apostrophes.
