Responding to "Do you mind..."

Do you answer "Yes..." or "No..." to show that you're willing to do what you're asked to do? Because I've heard both. Which one is correct?

Thanks. But I'm refering to a request starting with "Do you mind...?" For instance, if someone asks "Do you mind being my reference for a job interview?" to which you're willing to do. Do you respond with "Yes (meaning "I do mind?!"), of course" or "No (I don't mind...?!), of course"?

I assume you mean eg Do you mind opening the window?

No means I am willing, ie I do not mind.
Yes means I am unwilling, ie I do mind.

But we often do not give these short answers. Partly, I think, because they seem sometimes a

What the others said. But, as you know, sometimes you will get a yes or a no to the same question.

No, not at all, please sit down.

Do you mind if I sit here?
Yes, by all means, please sit down.

The first replies to the literal question. (Do I mind, no I don't).
The second replies to the underlying question. (May I sit down here?
