Yes, you need the word "of." There may be a few exceptions to this rule, but it is rare to use "regardless" without "of."

No, but the form without "of" is much more casual in style and is much less frequently used. If you check Google results, you'll find that "regardless of" is used 100 times more often than just "regardless". For this reason alone, I recommend keeping the "of".
