" the most tidy desk ? " or the tidiest desk

Can I say " which is the most tidiest desk here in the room ? " or should I say " which is the tidiest desk in the room ? "

You can say anything you want, but 'most -est' is incorrect grammar and redundant to boot. You should use either ' tidiest' or ' most tidy' , with the former the preferred.
