pronoun(it vs that and those vs them)

1. The biggest problem was the accent. That was difficult for me. (That = the accent)

Q) I was wondering if "that" is the only option. Can't a pronoun it go in place of "that?"
What's the difference between it and that?

2. There are a great number of people who are seeking employment, and a great number of those are married women.

You could in many cases. 'That' points in a direction, so we often use 'that' when the micro-topic is finished and 'this' when we are going to mention it again. 'It' doesn't do that, so it is a somewhat vaguer reference
