"You have been playing for three hours."
It means that for the last three hours you are playing and still you are playing. Right.
Now, can I say the same thing like this?
It has been three hours since you are playing.
Thanks in advance.

It has been three hours since you are playing.

"since", here, refers to the past. So, the playing started three hours ago and that fact should be expressed in the past tense: "It has been three hours since you started/began playing."
