I believe most native speakers will opt for 'on'. Unfortunately, there isn't a whole lot to be clarified though as it's essentially an idiomatic issue.

Do you mean "go" as in "turn"/"attempt"?

"on the first go" seems more feasible. If I wanted the "in" meaning, I think I would use "during".

However, more context is needed. It may be that "at the first attempt" is better.
