His hometown is the slums of Nigeria.

His hometown is in the slums of Nigeria.

Are both okay?

No. Slums are not a town, so you can't have "slums of Nigeria", and a town can't be in the slums because the slums are part of a town. You have to put it a different way. Maybe "He grew up in the slums of Lagos." or "His hometown is Lagos." I guess you could say "He grew up in Nigeria, in the slums.", but that is rather vague.
