"seems" is a verb inflection, not a plural.

Use "most seem ..." when "most" refers to something countable, e.g. "most (people) seem uninterested", and "most seems" when "most" refers to something uncountable, e.g. "most (of the money) seems to be missing". The same goes for agreement with other verbs.

When do you use "most seem" or "Most seems"?

If 'most' is being used as 'most people' (or most of some other countable entity), only 'seem' is possible.
If 'most' is being used with an uncountable noun, only 'seems' is possible.

'most' can also be an adverb (with either 'seem' or 'seems').
