Does the word, "Is" have to be capitalized when it is used in a title?

Yes, verbs are capitalized when using title case. Nouns and pronouns are capitalized as well.

Articles are not. Usually prepositions are not, though some style guides advocate capitalizing prepositions of a certain length. (I find that rule crazy and leave them all lower case.)
