Gosh, there are several alternatives (as noted by the fine folks who have replied). Here's another one: "Please reply as soon as you receive this message so I know that you received it."

Some other thoughts:
  • Personally, I would never write "reply back" because the word "reply" assumes that the recipient is to communicate back (i.e., reply) to the sender.

  • I thought I might simplify the sentence even further to "Please reply as soon as you get this message," but I caught myself! The sender wanted the recipient's reply so he/she (1) was

Re the first bullet point, the phrase "reply back" is redundant. One only needs to use the word "reply."

Re the second bullet point, sometimes editors edit TOO much. I was explaining why I didn't try to edit too much of the original sentence.
