There isn't a single, correct answer to this. Terminology used in education, as well as education systems, varies greatly in different parts of the world. For example, the term "high school" is used in some countries and territories for what is called "secondary school" in others.

In the US you'd hear:

"We have PE (or phys ed) in third period."

The school day in US high schools and junior high schools is divided into "periods": first period, second period, etc. So a student might have math in first period, history in second period, phys ed in third period, etc.

I don't agree with the "in" part.

I have gym (phys ed, PE) third period.
I have English fourth period.
I have physics fifth.
