In the section of "Synthesis and Transformation", if the given word is "only", should there be a comma in between?

For example:

Yiling took a much closer look at the thousand dollar note. then she realised that it was a counterfit.

Only when Yiling had taken a closer look at the thousand dollar note , did she realise that it was a counterfeit.
