1.He came on bus.

2.He came by bus.

Are bothe the sentences correct? IF yes, then what is diffrence between the two?

1.He came on bus. You need to say 'he came on the bus'.

2.He came by bus.

I'd say they are both correct.

#1 is more common in my experience. It also suggests that the speaker has some knowledge of the bus service.

#2 seems just a little bit more formal.
