Hi,

It depends on the sentence.

In some sentences, there would be no real difference.

eg

A: Would you like to have lunch next Tuesday?

B" Sorry, I'm busy (on) that day.

But consider a sentence like 'That day was the happiest of my life'. You can't use 'on' here.
