I wonder which one is more natural.

1. Two people of our staff attended the meeting.
2. Two people on our staff attended the meeting.

And is it better to use persons instead of people?

Only #2 is correct.

Two members of our staff

Two people on our staff

And is it better to use persons instead of people? 'People' is fine, and more natural.
