ok suppose if I want to instruct someone who is playing the game and he does not know how to operate it.

In the absence of more detailed instructions, it is presumed that the button will be displayed on the opening screen of the game, or will be in some other obvious place. If it is not in an obvious place then more detailed instructions will be needed, but just saying "on/in the game" does not actually add anything helpful.

Of course, you could use the last part of the same sentence to say something more useful and specific; for example, "In order to get the game started you need to click the play button on the opening screen".
