'Of' is necessary in writing, though it may not be spoken.

Thank you Mister Micawber,

I had 2 questions please if possible,

Do these 2 sentence have different meanings?

1 My parents are of two different religions.

2 My parents have 2 different religions.

And how would you personally phrase this sentence or would you phrase just the way I have?

1 My parents are of two different religions.

I would phrase them as I posted them. #2 is informal.
