now or by now?

hi guys! I need your help, a native's sense!!
I want to know if these sentences below make sense.

Jane left for her hometown early morning. His family is waiting for over an hour.

Jane should be here by now.
Jane should be here now.

The bus should have arrived by now.
The bus should have arrived now.

These two pairs of sentences means the same? or Either of them is correct?
If both of them are correct, please tell me the difference.

Lastly, Surfing the web, I saw the second one below(with 'now' , not 'by now') somewhere. Similarly I wonder if these two means the same or either of them is correct.

The window should have popped up automatically by now.
