Not slightly/not a little

1-They were not slightly interested by his offer.
2-They were not a little interested by his offer.

3-They were not slightly affected by their defeat.
4-They were not a little affected by their defeat.

In which cases:
a-They were not interested/affected at all
and in which cases
b-They were interested/affected a lot
